Project: Subcellular innervation pattern of somatostatin positive (SST+) axonal boutons in the mouse barrel cortex

A study on the connectivity of SST+ interneurons with other types of neurons in the mouse barrel cortex. Neocortical SST+ interneurons are diverse and locate in layers 2 to 6. Distinctive subtypes of SST+ neurons have complementary axonal distributions and different microcircuits, with regard to layers (Oliva et al. 2000; Ma et al. 2006; Xu et al. 2013; Muñoz et al. 2017). It is important to quantitatively know their axonal innervation patterns, in order to understand their structure-function relationships in sensory information processing.

Due to the superiority in resolution, electron microscope (EM) is a great tool to visualize the ultrastructure of a synapse. Utilizing electron dense immunostaining, it is possible to study the synaptic connectivity of neurons at the subcellular level. The main work is to do postembedding anti-GABA immunogold labeling on serial ultrathin sections (60 nm thick) and quantitatively analyze the postsynaptic targets of SST+ axonal boutons under the EM.
